I have the basics for a story idea I might work on in novel form. So far I have main characters and most of the backstory worked out in my head. Although I haven't decided where to go with it exactly. I'm working on character design but I'm not sure if I want to try to draw them as I'm not a great artist.
Main characters are twin loli girl vampires with twintails aprox 12yo in appearance.

Betrayal is a main theme, either each other or family (childhood friend) i'm a fan of prophecy or Legend in the making. Imagine pulling out the sword noone else could? (sound familiar?)
Sci Fi setting has been on my watch list and if done right can be amazing.
O.o maybe a futuristic city is based around a location where mythological weapons and items are sealed in plain sight and everyone comes from all over the world to pull one out. Your characters can pull out whatever you can imagine, be it a Twin sword for each girl or pistols/nunchuks XD
Maybe the items have personalities? Maybe the weapons entail Royalty once removed?

You know, I've had an idea in my head for a long ass time. Like you, I had the general characters, setting, and themes decided, with intricate backstories for everyone and every thing in the town. And also like you, no idea with where to go with the story. This is what's becoming of it. (I haven't run a PR campaign yet, so don't worry about feeling obligated to donate or anything. I'm stable at the moment.)

Seriously, if you aren't confident in your artistic abilities, its worth it to save up to commission a professional artist to draw your characters. And keep those images with you as you develop your world, story, and characters, to get to a point where you could talk to a professional writer to help you pan out a story out of them. An idea is a dime a dozen. A visual representation in conjunction with an idea can get you much further when looking for people you need to help you achieve your goals.
By the way, as someone with an identical twin brother, I could tell you how to make twins in a more relatable manner, if you ever want an idea bouncer.

What makes a good story is originality. Make something that will be thought provoking and is not usual. Don't go overboard because people can grow to dislike the oddness, but go with something that fits well. Study types of animes and mangas you like, understand the storyboard of them and look at your characters. Look at their beginning and their weakness and see where you want them to grow so that they can become stronger. If you are not good at art, I'd recommend asking a friend or finding someone to help. Granted, make sure they sign a non-disclosure agreement or have written say that they will not steal your work or idea.

I am currently making one. I am drawing the scenes already. My story is about a guy who was turned into stone by a curse. Over 5,000 years later the curse is broken by a local girl and the guy looks around confused. This is the start of his destiny. It's a magic, demon, romance, light comedy, action and adventure. I post a novel form of the first two chapters, but I am still working on those chapters.

Here's my advise: go for it!
That is how I started writing fanfiction, I just went "screw it" and wrote it. I'm not famous of course, but I'm a lot happier than I was "back then."
Anyway, go for it! Don't sit back thinking your not good enough to do this, because I bet you are.

Writing 101.
Fuck your character backstory.
Fuck your main character.
Plot appeal is what's important. Not even genre, or style or arc or concepts. PLOT must be rock solid.
Supporting cast over main characters, the biggest bad habit of new writers is writing supporting cast as plot devices. Don't. The story is interesting because of the situations the main characters find themselves. What defines a main character is how they are defined by these situations. Not how the main character handles them.

You and me both Loli_Vampire. I am also writing a story that I hope to turn into a manga, but so far due to my inability to draw anything that isn't realistic I am stuck with doing novels until I can find an artist willing to do it. Only difference is mine is about a mental and physical war between a man and his id (the part of the mind that represents evil and primal thought) and how it eventually leads to him attempting a world-wide dictatorship for the sake of peace.
