Being just a normal student without a special talent, Makoto Naegi wins a lottery to attend the prestigious Hope's Peak Academy where only the top prodigies attend. However, instead of this being the beginning of a wonderful high school life, it's a ticket to despair, because the only way to graduate from Hope's Peak Academy is to kill one of your fellow students or be one of their victims.

Hope's Peak Academy is a special school, recognized by the government, that was established in order to gather and nurture top-class students who excel in various fields. This school has two courses: The main course, and the preparatory course.

The world was close to being destroyed by Ultimate Despair, who caused "The Biggest, Most Awful, Most Tragic Event in Human History." In order to save this world from the event's chaos, the Future Foundation was formed.

In 2012, the world allegedly comes to an end at the hands of a "human-made" virus, ravaging the global populace and leaving only children under the age of thirteen untouched. It is at this time that vampires emerge from the recesses of the earth, likely followed by age-old horrors of the dark thought only to be myth.

Yuuichirou Hyakuya is finally reunited with his childhood friend Mikaela Hyakuya, whom he had long presumed to be dead. Upon their reunion, however, he discovers that Mikaela has been turned into a vampire.

In the year 2021, mankind was defeated by Gastrea, a parasitical virus, and is forced to live within a wall made of Varanium Monoliths, a metal that is able to subdue Gastrea. Soon, the "Cursed Children", children born with the Gastrea virus who are able to control it—which gives them superhuman abilities—were discovered.
